ABSTRACT

Leukemia inhibitory factor (LIF) contributes to the neuroprotection by neural stem cells (NSCs) after ischemic stroke. Our aim was to explore whether LIF-transfected NSCs (LIF-NSCs) can ameliorate brain injury and promote neuroprotection in a rat model of cerebral ischemia. To accomplish this goal, we transfected NSCs with a lentivirus carrying the LIF gene to stably overexpress LIF. The LIF-NSCs reduced caspase 3 activation under conditions of oxygen-glucose deprivation in vitro. Transient cerebral ischemia was induced in rats by 2 h of middle cerebral artery occlusion (MCAo), and LIF-NSCs were intravenously injected at 6 h post-ischemia. LIF-NSC treatment reduced the infarction volume and improved neurological recovery. Moreover, LIF-NSCs improved glial cell regeneration and ameliorated white matter injury in the MCAo rats. The NSCs acted as carriers and increased the expression of LIF in the lesions to protect against cerebral infarction, suggesting that LIF-NSCs could be a potential treatment for cerebral infarction.

ABSTRACT

Lysozyme, pepsin, ovalbumin, hemoglobin andγ-globumin were chosen as templates to investigate the imprinting capability of amphoteric polyacrylamide cryogels. Prepolymerizing solutions contained acrylic acid and allyl amine, as well as acrylamide and N,N'-methylenebisacrylamide as the functional monomers. As a result there were both acidic and basic functional groups in the polymers, facilitating effective interactions with likewise amphoteric proteins. The proteins differ greatly and cover wide scopes of molecular weights and isoelectric points. Regardless of the values of the molecular weights and isoelectric points, all the templates gave higher retentions on the MIP tubes than on the NIP tube. The MIP of lysozyme indicated the highest imprinting factor of 7. 0, and that of γ-globumin showed the lowest, 2. 0. The values of other proteins were intervenient. Conclusively the amphoteric polyacrylamide cryogels were suitable imprinting materials for various proteins, and could potentially be used for protein recognition, purification, and depletion.

ABSTRACT

Cryogel is a kind of elastic, spongy hydrogel prepared via addition polymerization from monomers or cross-linking reaction from a polymer at a temperature lower than the freezing point of the solvent. Cryogel has been principally used in molecular imprinting cryogel based solid phase extraction in the field of analytical chemistry, which combines both the advantages of high selectivity of the molecular imprinting and the porous structure, high throughput of a cryogel. Molecular imprinting cryogel can be used as the stationary phase in a packed column, a monolith column, or the adsorbent in a separation membrane. A variety of molecules can act as template molecules, including small molecules, macromolecules, a certain fragment of a macromolecule, and even an inorganic ion. Both a simple template and a complex template can be used. When employing more than a molecule ( or an ion) as templates simultaneously, the method is termed as Mixed template. While high abundance components of a complex sample act directly as mixed template, we name the method as Pending template. Further, while an analogue, an isotopic label, or a fragment of the target substance acts as a template, the method is defined as Dummy template. However, the research on cryogel is insufficient and the application of the cryogel is narrow hitherto.

ABSTRACT

Background Graft rejection is a primary cause of corneal transplantation failure, especially in high-risk keratoplasty. How to extend the survival time of graft is a problem to be solved. Objective This study was to investigate the influence of immune tolerance on high-risk rat keratoplasty induced by donor bone marrowderived immature dendritic cells (imDCs) transfected by chemokine receptor 7 (CCR7) recombinant adenovirus (Ad).Methods Bone marrow-derived imDCs were isolated and cultured from femur marrow of one male Wistar donor rat.The cells were transfected using recombinant Ad vector with rat CCR7 gene and resuspended in 500 μl PBS containing 1% fetal bovine serum with the cells 1 × 107.High-risk corneal transplantaion models were established using monolateral corneal alkali-burn method in 60 SD rat recipients, and then allograft keratoplasty was performed with the 30 Wistar rats as donors. The models were randomized into the PBS group,imDCs group,imDCs with blank Ad vector group and imDCs with Ad-CCR7 group following the corresponding solution injection via caudal vein on preoperative day 7 and postoperative day 3 respectively. The survival time of graft was evaluated under the slit lamp microscope once per day.On the 14th day after operation, corneal sections were prepared from 6 eyes of each group for the pathological examination, and the relative expression levels of T helper cell 1 (Th1)-related factors,interleukin-2 (IL-2) mRNA and interferon-γ(IFN-γ) mRNA,as well as Th2-related factors, IL-4 mRNA and IL-10 mRNA, were detected by reverse transcription PCR (RT-PCR).The use and care of animals complied with the ARVO statement Results The mean survival time of grafts was (10.44±1.88) , (16.00±2.18) , (15.11±2.03) and(23.67±2.83) days in the PBS group,imDCs group,imDCs with blank Ad group and imDCs with Ad-CCR7 group, respectively, with a significant difference among the 4 groups (F =53.005, P =0.000).Compared with the PBS group, the mean survival time of grafts was considerably extended in the imDCs group,imDCs with blank Ad group and imDCs with Ad-CCR7 group (t=5.220,4.385,12.423 ,all at P=0.000) ,and a remarkble prolongation of graft survival duration was seen in the imDCs with Ad-CCR7 group in comparison with the imDCs group and the imDCs with blank Ad group (t =7.204,8.039,both at P=0.000).The relative expression levels of IFN-γ mRNA and IL-2 mRNA in the grafts were significantly lower, but the relative expression levels of IL-4 mRNA and IL-10 mRNA were significantly higher in the imDCs group,imDCs with blank Ad group and the imDCs with Ad-CCR7 group than those in the PBS group (all at P<0.05).Compared with the imDCs group and the imDCs with blank Ad group, the relative expression levels of IFN-γ mRNA and IL-2 mRNA in the grafts were remarkably delined, but the relative expression levels of IL-4 mRNA and IL-10 mRNA were remarkably elevated in the imDCs with Ad-CCR7 group (all at P =0.000).Conclusions Application of imDCs transfected with CCR7 recombinant Ad via vena caudalis can prolong the survival time of grafts after keratoplasty of SD rats probably by affecting the balance of Th1/Th2 cytokines.

ABSTRACT

Objective To establish and validate a modified rat thromboembolic stroke model. Methods After taking femoral arterial blood and mixing it with thrombin, they were injected into PE-50 catheter for preparing in vitro thrombosis in 60 Sprague-Dawley rats.A thromboembolic cerebral ischemia model induced by catheterization of the right external carotid artery and the small blood clot emboli were injected into the internal carotid arteries.Thirty rats were randomly divided into a large number of emboli group (n =10 with 12 emboli),a median number of emboli group (n =10 with 10 emboli) and a small number of emboli group (n =10 with 8 emboli). Two hours after embolus injection, the neurological deficit score was performed and the success rate of the model was compared in all groups.Twenty-four hours after embolus injection, the rats were sacrificed and the brains were removed for 2,3,5-triphenyltetrazolium chloride staining. The hemorrhage, infarct volume, bleeding incidence and mortality after cerebral infarction were evaluated. The high success rates of the modeling in the emboli groups were selected and they were randomly divided into either a normal saline group (n =12) or a recombinant tissue-type plasminogen activator (rtPA) group (n =12).The rats were given normal saline and rtPA at 3 hours after embolus injection. Before embolus injection and 2,6,12 and 24 hours after embolus injection, the neurological scores were performed respectively; 24 hours after embolus injection, the rats were sacrificed and the brains were removed for 2,3,5-triphenyltetrazolium chloride staining. The hemorrhage rate, infarction size, degree of cerebral edema, and blood-brain barrier permeability were evaluated. Results Only 40% of rats had neurological deficits in the small number of emboli group, and the infarct volume was only 10.54 ± 2.82%.The success rates in the median and large number of emboli groups were 80% and 100% respectively. They were all significantly higher than those in the small number of emboli group (P =0.011 ).The infarct volume was also significantly greater than that in the small number of emboli group (F =40.897,P =0.000).After administration of rtPA,the mean survival time of the rats in the large number of emboli group was less than 24 hours,so the median number of emboli group was selected to study the thrombolytic effect of rtPA.The infarct volume and neurological function score in the rtPA group were improved significantly compared to the normal saline group (t =7.728,P =0.000),while there were no significant differences in the hemorrhage rate, degree of brain edema and blood-brain barrier permeability between the 2 groups. Conclusions The stability and reproducibility were good in the modified thromboembolic cerebral ischemia model injected with 10 emboli, the neurological function was improved significantly after thrombolysis, and it was applicable to the experimental study of pathophysiology of cerebral ischemia and thrombolytic therapy.

ABSTRACT

Objective To describe the epidemiological features of severe cases and deaths of pandemic influenza A (H1N1) 2009 in Beijing and to explore the main risk factors associated with the disease severity. Methods Data of pandemic influenza A (H1N1) 2009 reported in Beijing were collected and analyzed. The relative risks between disease condition and possible risk factors were determined using the multi-factor Logistic regression analysis. Results The incidence of pandemic influenza A(H1N1) 2009 was 66. 1/100 000, and the highest incidence was 86. 8/100 000 which occurred among people aged 25-60 years old. The prevalence of critical patients (12. 5/100 000 and 3.9/100 000, respectively), mortality (0. 9/100 000 and 0. 7/100 000, respectively) and fatality (2.4 % and 3.3 %, respectively) in people aged 0-5 years old and above 60 years old were higher than other age groups. There were 110 (20. 0%) preschool children among 549 critical cases, which were the largest proportion of critical cases. Among 69 death cases, 17 (24. 6%) were retirees with the highest proportion of death cases. More than 70. 0% of the critical cases and deaths visited hospital within two days of onset. The chronic heart disease and chronic lung disease were the most frequent risk factors of severe and death cases. Multi-factor Logistic regression analysis showed that the variables including aged above 60 years old (OR = 3. 586, 95 % CI = 1. 586 - 8. 117), chronic heart disease (OR=2. 126, 95%CI= 1. 178-3. 835), and chronic lung disease (OR=1. 954, 95%CI=1. 126-3. 391) were significantly associated with the disease severity. Conclusion Factors of aged above 60 years older, chronic heart disease and chronic lung disease may aggravate the severity of influenza A (H1N1).

ABSTRACT

BACKGROUND: Early education is an important step in raising children. Many facts prove that early education can promote infants' intellectual development so as to lay good foundation for their future development.OBJECTIVE: To understand the status of infants' intellectual development with Wechsler Intelligence Scale for Children and analyze the effects of early education.DESIGN: Comparative observation.SETTING: Wards of the Department of Pediatrics and Department of Obstetrics, Xuanwu Hospital of Capital University of Medical Sciences.PARTICIPANTS: We recruited 100 normal newborn infants born in the Department of Obstetrics, Xuanwu Hospital of Capital University of Medical Sciences, between January 2000 and December 2001. Guardians of the infants were informed of the research objective. Infants were divided into 2 groups according to their birth date: early education group (n=50) in which the infants were born on Monday, Wednesday, Friday or Sunday; control group (n=50) in which the infants were bom on Tuesday, Thursday or Saturday.METHODS: ① All the parents of the infants in the two groups received the guidance of feeding, infection prevention, inoculation and other health care guidance. On this basis, the parents of the infants in early education group were informed of intellectual development disciplinarian and educational guideline for 0-3 year-old infants when physical examination was performed on the infants at newborn period (twice) and 42 days after birth (once). The parents were asked to raise their infant according to the requirements of the guideline and communicate with the physicians at any moment. In control group, routine physical examination was performed on the infants at new-born period (twice) and 42 days after birth (once), and their parents were not given the early education of pediatric system. ② At 4 months and 3 days to 4 months and 29 days after birth, the infants in the two groups were taken back to hospital. Status of intellectual development was evaluated with intelligence scale for children revised and standardized by the Institute of Psychology of the Chinese Academy of Sciences and China Children Development Center according to Bayley's Scales of Infant Development [It consisted of intellectual scale and movement scale. Intellectual scale was used for 4.0-5.3 months babies (4 items, Item E: reaching out for building blocks, hand-eye coordination when reaching out, pick up building blocks, and keeping reaching out for things; Item G: noticed small pills; Item H: approached ocular; Item C: fond of playing with him).Movement scale was used for 4.0-5.3 months babies (4 items, Item A: turning their heads freely; Item E: from lying on the back to lying on the side;Item D: part of pollex face to face when holding building blocks; Item C:could sit independently for a moment)]. The primary scores of the infant was obtained from the number of an item which the infant could complete in intellectual development scale, then intellectual index and psychological movement development index were obtained according to his age span from the scale. Those with scores >130 were considered excellent, and those with scores < 69 were mental retardation. ③ t test and U test were used in comparison of the differences in metrical and quantitative data.MAIN OUTCOME MEASURES: ① Comparison of intellectual development index and psychological development index between the infants in the two groups. ② Status of infants' intellectual scale and movement scale completion.RESULTS: Totally 100 infants entered the result analysis. ① Intellectual development: Intellectual development index and psychological movement development index were significantly higher in early education group than in control group (103.94±7.64, 111.20±9.40; 93.92±5.18,99.22±5.52, t=8.3,4.7, P < 0.01). ② Status of intellectual scale completion: The number of infants who could complete Items E, D and C was obviously greater in early education group than in control group [36 infants (72%), 8 infants (16%), u=5.6, P < 0.01]. ③ Status of movement scale completion: The number of infants who could complete Items E, D and C was obviously greater in early education group than in control group [45 in fants (90%), 32 infants (64%), 48 infants (96%); 30 infants (60%), 12 infants (24%), 42 infants (84%), u=2.0-4.89, P < 0.05-0.01].CONCLUSION: Early education can improve infants' intellectual development, especially manipulative ability.

Geometric models were built by the use of advanced 3-dimensional laser scanning and computer assisted 3-dimensional design technique. The geometric data were introduced into the finite element software for establishing the finite element mesh after being processed. Results:The 3-dimensional finite element models of the minimum distal occlusal structure defect with restoration were created, which included metal crown with exterior anatomic morphology of the mandibular first molar, tooth preparation, filling material and the structure of periodontal membrane. Conclusion: The method may be a useful approach for building models of teeth and relative tissues.
